中文摘要: 近年研究证明骨髓间充质干细胞能够显著缩小心肌梗塞面积,促进心功能恢复,将为该病的治疗带来重大突破。但由于骨髓间充质干细胞远程移行能力低,影响其静脉使用效果,目前需采用直接心肌内注射。因此,提高骨髓间充质干细胞的移行能力对于提高治疗效果、方便临床使用至关重要。粘合蛋白βV 是存在于移动活跃的细胞上的表面受体,我们发现有1/3 的骨髓间充质干细胞也表达该受体,而且其配体vitronectin 在心肌梗塞后表达显著增加。我们设想粘合蛋白βV 是影响骨髓间充质干细胞向梗塞心肌移行的重要受体。本课题将通过细胞分选技术筛选出粘合蛋白βaV+和粘合蛋白βaV-的两种间充质干细胞,然后通过对其在粘附、存活、细胞活性因子释放、局部移行和静脉注射后向梗塞心肌远程定向移行及对梗塞心脏修复能力的比较研究,明确表达粘合蛋白βV 的骨髓间充质干细胞是否具有较强的移行能力和较好的治疗效果。

英文摘要: Bone marrow derived mesenchymal stem cells (MSCs) have been shown to improve myocardial infarct and ventricular performance, and demonstrated significant potential for the treatment of myocardial infarction. However, the ability of homing of MSCs, particularly after ex vivo expansion, decreases, and decreased homing efficiency of MSCs reduce their therapeutic effect after infusion. Integrins aVb3 express in cells with active ability of migration. In our preliminary study, we found that a third of MSCs expressed integrins aVb3, and their ligand vitronection was upregulated in the ischemic myocardium. We hypothesize that integrins aVb3 are important for the homing of MSCs to the infarcted myocardium. In this study, we will isolate MSCs expressing integrins aVb3, and compare their abilities in adhesion, migration, survival, cytokine release and homing to the infarcted myocardium with MSCs negative for integrins aVb3, thus determining their involvement in MSC homing to the ischemic myocardium.
