中文摘要: 恶性肿瘤是严重威胁人类健康和生命的重大疾病之一,恶性肿瘤的早期发现、早期诊断是目前延长癌症患者生命的最有效手段之一。恶性肿瘤的发病与特定生物标志物在体内的表达量存在密切的关系,因此肿瘤标志物高灵敏度和特异性的检测对肿瘤的早期筛查、临床诊疗具有十分重要的意义。纳米探针作为新型的功能探针,具有体积小、容易进行生物标记等诸多特点,已广泛应用于药物筛选、疾病诊断、细胞及活体成像等领域。本项目拟通过制备AuNPs@MoS2纳米复合材料,构建多种AuNPs@MoS2纳米探针,结合电化学分析技术快速、灵敏度高、稳定性好的优点,构建电化学免疫传感器。利用纳米探针的信号放大作用,实现肿瘤标志物的超高灵敏和特异性检测。通过对肺癌标志物CEA、NSE等的联合检测,提高肿瘤早期筛查的准确性,为纳米电化学生物传感器应用于癌症早期检测和诊断提供新的纳米材料和新的科学思路。

英文摘要: Malignant tumor (cancer) is one of the terrible diseases of human being, which seriously threatens human health and life. Detection of cancer at the early stage is one of the important means to prolong the life of patients. Therefore, it is significant to detect tumor biomarkers with high sensitivity and specificity for clinical diagnosis and treatment. Nanoprobe is considered as a new functionalized probe with a small size and easily functionalized with biological markers, which has been widely applied in drug analysis, disease diagnosis, cellular and in vivo imaging and so on. The project designs kinds of AuNPs@MoS2-based nanoprobes to construct electrochemical immunosensors for tumor biomarkers detection with high sensitivity and specificity. The immunosensor combined with the advantages between AuNPs@MoS2 nanomaterials and electrochemical analysis technique, which can be used to simultaneously detect several lung cancer biomarkers. The novel AuNPs@MoS2-based immunosensor maybe offers an opportunity for applying in cancer diagnosis.
