中文摘要: 苹果多酚具有降血脂、预防心脑血管疾病的功能。根皮苷是苹果多酚中的特征多酚类活性物质,本项目以仓鼠为动物模型,通过根皮苷对高脂高胆固醇膳食喂饲仓鼠体内胆固醇代谢平衡的研究及对血脂水平、体内胆固醇积累水平、胆固醇代谢排出水平的影响,系统评价根皮苷的调控体内胆固醇代谢作用。并采用荧光标记、放射性同位素标记等测试手段和Real Time-PCR、Western Blot等研究方法探讨根皮苷对调节胆固醇小肠吸收(ACAT)、低密度脂蛋白胆固醇肝脏吸收(LDL-R)、内生合成(HMG-CoA还原酶)、肝脏胆汁酸代谢(LXR、CYP7A1)、血液胆固醇酯转移(CETP)等胆固醇体内不同代谢途径和各途径中相关关键酶的活性、蛋白、基因表达的影响。课题的研究将在整体上阐明根皮苷调控机体胆固醇代谢的分子作用机制。

英文摘要: Previous reports demonstrated that consumption of apple polyphenol had been associated inversely with cardiovascular diseases and hypercholesterolemia. Phloridzin is a special of phenolic compound in apple. This item is plan to investigate the effect of phoridzin on cholesterol blance in hamsters and blood cholesterol level, tissue cholesterol level, cholesterol excretion in Golden Syrian hamsters maintained on a high fat and high cholesterol diet. With the methods of fluorescence labeling, radioactively labeled isotope, Real Time-PCR and Western Blot technologies, the activity, protein expression and gene expression of cholesterol-regulating enzymes will be determined including intestinal cholesterol absorption(ACAT),hepatic LDL cholesterol uptake(LDL-R), hepatic cholesterol de novosynthesis(HMG-CoA Reductase), hepatic conversion to bile acids(LXR、CYP7A1)and cholesteryl ester transfer between lipoprotein classes in the blood(CETP) in Golden Syrian hamsters when hamsters are fed a diet supplemented with phloridzin. And the results will explain the hypocholesterolemic activity of phloridzin and its underlying mechanism.
