中文摘要: 三阴性乳腺癌(TNBC)是一类恶性度高、预后差的乳腺癌亚型,转移是影响其预后的主要因素。我们前期工作发现:TNBC中富含肿瘤干细胞(CSC),并且CSC显著影响了TNBC的生物学特征与临床预后;TNBC患者体内存在少数循环肿瘤细胞(CTC),也同样具有CSC特性。在此基础上提出设想,干细胞样循环肿瘤细胞(S-CTC)是促进TNBC转移的根源,并且具有独特的遗传特征。为验证该设想,我们拟采用专利技术(CanPatrol系统)检测并分离TNBC患者体内的S-CTC,通过单细胞技术研究其遗传特征,从遗传学和临床水平分别证实TNBC转移中S-CTC的作用,并绘制S-CTC特征性遗传变异图谱。为进一步研究TNBC及其转移的特征性诊断、靶向性干预或者准确性预后评价,提供理论基础与候选靶点。

英文摘要: Triple-negative breast cancer (TNBC) represents a subtype of breast cancer with high aggressivity and poor prognosis. Metastasis is one of the main prognositic factors. In our preliminary study, we found that cancer stem cells (CSC) were enriched in such subtype. The biological characteristic and clinical prognosis were associated with the CSC in TNBC. We also found some circulating tumor cells (CTC) representing the feature of CSC. We have a hypothesis that stem-like circulating tumor cells (S-CTC) promote the metastasis in TNBC, with unique genetic characteristics. In the study, S-CTC will be detected and separated from TNBC patients by CanPatrol system. The genetic features of S-CTC will be studied by single cell sequencing. The role of S-CTC in metastasis will be verified in a genetic study and a clinical trial. The special genetic variation of S-CTC can also be detected in the study. Such research will be helpful for the deep study on TNBC and its metastasis.
