中文摘要: 肾虚证是糖尿病的重要基础证型,前期糖尿病肾虚证家系的差异表达基因集中于转录调控、免疫和代谢类基因模块。长链非编码RNA可通过多种方式调控相关基因的表达,因而引入lncRNA研究,探索前期结果的机制。精选6个典型的肾虚证糖尿病家系,将患者和正常人进行lncRNA芯片实验,筛选与糖尿病肾虚证相关的候选差异lncRNA,预测差异表达lncRNA潜在靶标,采用Western及RT-PCR的方法鉴定lncRNA是否通过降解靶标mRNA或抑制蛋白翻译起作用,检测糖尿病肾虚患者及正常人对照潜在靶标的mRNA及蛋白表达水平。本研究将采用RNA组学方法,结合生物信息学方法,建立高通量筛选和鉴定lncRNA技术平台。在中医证候的基因组研究平台上,构建以lncRNA为核心的基因表达及蛋白质调控网络,以解析lncRNA在糖尿病肾虚证发生和发展的作用机制。

英文摘要: Preliminary survey showed that diabetes is connected with kidney deficiency syndrome. Use of mRNA microarray, some metabolism, transcriptional regulation and immune genes were screened out which may play important roles in type 2 diabetes with kidney deficiency syndrome. Long noncoding RNAs (lncRNAs) are an important class of pervasive genes involved in a variety of biological functions. 6 diabetic with kidney deficiency pedigrees will be elected, divided into healthy , divided into three groups of healthy , diabetic with kidney deficiency patients and diabetic with non-kidney deficiency patients. Use of lncRNA microarray, verified by differentially expressed mRNA and protein expression experiments, some candidate differentially expressed lncRNA can be screened out which may play important roles in type 2 diabetes. This study will aim to explain the lncRNA regulatory mechanism in diabetes with kidney deficiency pedigree.
