中文摘要: 磁性纳米材料因其独特物理化学性质和广阔生物医学应用而引起研究者的广泛关注。高温热解法制备的磁性纳米粒子具有结晶良好,粒径均一,磁响应性好,粒径单分散等性质而成为磁性纳米粒子制备的主要方法,但该方法制备的磁性纳米粒子表面往往吸附有稳定剂油酸,这使其在水相中不能分散。因此将单分散磁性氧化铁纳米粒子的水相转成为近年研究的热点。本项目提出利用磁性纳米材料表面油酸的可控氧化反应,建立一种单分散磁性纳米粒子水相转移的新方法。拟通过系统优化氧化反应条件,在控制氧化反应动力学的同时,研究氧化过程对氧化铁纳米粒子的组成、晶型、大小、磁学等的物理化学性质影响,探讨氧化剂对氧化铁纳米粒子作用的过程和机制,从而得到功能团可控的水相转移方法。再利用生物大分子偶联,体外细胞毒理学研究及药物负载与释放等实验,评价其在生物分子偶联效率、细胞安全性及磁靶向载体的性能,从而为其将来在生物医学领域诊疗中的应用奠定基础。

英文摘要: Iron oxide nanoparticles (NPs) have been extensively studied for biomedical applications, such as in targeted delivery, in localized therapy, and as contrast agents for magnetic resonance imaging (MRI). The water solubility and surface functionality of NPs are key parameters to their interactions with biological systems. Currently, high quality iron oxide NPs regarding monodispersity, size distribution, and crystallinity are normally produced in organic solvents and stabilized with oleic acid (OA). Therefore, phase transfer to achieve NP water solubility and functionality for further conjugation is essential and remains an outstanding challenge. Herein, a novel method is proposed to transfer monodisperse iron oxide nanoparticles to water based on the controlable oxidation of OA coated on the surface of magnetic particles. the oxidation can be controlled by systemic optimzation including the oxidatives, solvents, temperatures to obtain the magnetic particles functionalized with epoxyl, hydroxyl, carbonyl and carboxyl groups. Based on the oxidation effect on the properties of iron oxide nanoparticles during the process of transfer, the interaction between the oxidative and magnetic particles is explored.
