中文摘要: 创伤后应激障碍(Posttraumatic stress disorder, PTSD)是个体暴露在异乎寻常的威胁性或灾难性应激事件后出现的一种持续而严重的心理疾患。遗传因素在PTSD的发病过程中起到重要作用。人类和动物的研究显示催产素及其受体通路可能是个体经历严重创伤性事件后,出现异常心理与生物应激反应的重要介体,提示其在PTSD的发生和发展中可能具有重要的作用,但是目前尚无研究系统而深入地探讨二者的关系。本研究拟依托我单位在汶川地震后建立的研究基地与样本库,采用多种研究范式与手段,探讨催产素的血液水平、催产素及其受体通路的基因变异以及甲基化水平与PTSD症状以及相关的异常行为学和生物学指征之间的关系。本研究的发现不仅在理论上有助于阐释催产素及其受体系统在PTSD发病机制中的作用,促进对PTSD发病机制的理解,同时在临床实践中有助于鉴别可供临床使用的生物标记与治疗靶点。

英文摘要: Posttraumatic stress disorder (PTSD) is a complex and severe mental disorder triggered by exposure to an extraordinarily traumatic event. Many studies have provided evidence supporting a genetic predisposition to PTSD. Human and animal studies suggest that the oxytocin-oxytocin receptor system may be an important mediators of abnormal stress responses following exposure to traumatic events, which implicates an important role of this system in the development of PTSD. In the present proposal, we aim to systematically investigate the associations between the oxytocin-oxytocin receptor system and PTSD on the basis of research sites and databases established by our institute after the Wenchuan earthquake. By using molecular genetics, epigenetics and molecular biological techniques, we will analyse blood levels of oxytocin, and genetic variation and methylation of the oxytocin-oxytocin receptor system, and further examine the associations between these indicators and PTSD symptomatology, and PTSD related abnormal behavorial and biological indicators. Findings from the series of studies are pertinent for further understanding of the underlying mechnism of PTSD, and may help to identify novel biomakers and potential therapeutic targets for PTSD.
