中文摘要: 酒酒球菌 (Oenococcus oeni, O. oeni) 是葡萄酒苹果酸-乳酸发酵(Malolactic fermentation, MLF)的主要启动者,在葡萄酒酿造工业中的重要地位。研究通过利用高通量测序技术获得Oenococcus oeni SD-2a混合样本的转录组信息,并对其进行生物信息学分析。再利用升级版的DGE(Up-graded Digital Gene Expression)技术分析O.oeni SD-2a的抗逆基因的表达特征。由于抗逆相关基因在O.oeni SD-2a的抗酸和抗酒精过程中起关键性作用,实验将同时利用模式生物大肠杆菌验证O.oeni SD-2a的抗逆基因hsp 18基因和clp基因家族在抗逆过程中的功能。研究旨在揭示酒酒球菌胁迫条件下的抗逆基因表达特性,从而提高其在环境胁迫下的生存能力和活力,同时为直投式活性乳酸菌发酵剂的发酵效率提供了理论依据。

英文摘要: Oenococcus oeni is the main start Wine of malolactic fermentation (Malolactic fermentation, MLF). It plays an important role in the Wine brewing industry. The study obtained the transcriptome information from Oenococcus oeni SD-2a mixed samples by using high-throughput sequencing technology, and analyze it with the bioinformatics. The DGE (Up-graded Digital Gene Expression) technique is used to analyze the expression of resistance gene of O.oeni SD-2a. Because the resistance genes play a key role in the O.oeni SD-2a acid and alcohol resistance process, the study will use model organism Escherichia coli to verify of the stress resistance function of resistance gene hsp 18 and clp family in O.oeni SD-2a. The aim of the study is to understand the resistance gene expression characteristics of Oenococcus oeni under the stress conditions, so as to improve the environmental stress in the viability and vigor, and provides the theory basis for the fermentation efficiency DVS lactic acid bacteria fermentation agent.
