中文摘要: 蛋白质生物标志物的检测,在癌症等重大疾病的预警和诊断方面具有极其重要的作用。对癌症标志物进行准确快速的测定,能够为癌症的早期诊断和临床治疗提供重要且及时的依据,从而有效提高癌症患者的存活率。但是在生物样品中,大多数癌症标志物的浓度都极低,而且与众多其它的蛋白质共存,因此对它们的测定需要有极高的灵敏度和选择性。本研究计划致力于发展一种基于单根导电聚合物纳米线的适体传感器,用于超灵敏地测定复杂生物样品中癌症相关的蛋白质标志物。我们通过简化的模板设计,采用电化学方法可控地原位合成单根的导电聚合物纳米线,然后在纳米线上修饰适体,用于检测与适体相结合的癌症相关蛋白质标志物。该纳米生物传感器综合了单根导电聚合物纳米线的易修饰性和超高灵敏性,以及适体的高选择性,从而有望在复杂的生物样品如血液中选择性的检测出极其微量的癌症标志物。

英文摘要: The detection of protein biomarkers plays an essential role in the precaution and diagnosis of important diseases such as cancer. The fast and accurate detection of cancer biomarkers can provide important and timely support for the early diagnosis and clinic treatment of cancer, and thus improve the survival rate. However, in biological samples, the concentration of cancer biomarkers are normally very low, and in addition, they are co-existed with a lot of other proteins. As a result, the detection of cancer biomarkers need to be highly sensitive and selective. In this proposed research, we will develop an aptasensor based on a single conducting polymer nanowire for the ultrasensitive detection of cancer biomarkers in complex media. Through a simplified template design, we can electrochemically synthesize single conducting polymer nanowire in situ in a controllable way. After the modification with proper aptamer, the aptamer-functionalized single nanowire can be used for the detection of the corresponding cancer biomarker. The proposed nano-biosensor combines the unique properties of single conducting polymer nanowire(easy for modification and highly sensitive) and aptamer (highly selective), and therefore it can be used for the selective detection of trace amount of cancer biomarkers in complex biological samp
