中文摘要: 基因组测序技术已趋成熟,并且测序费用已大幅度降低,基因组测序将成为食用菌分子生物学研究的基础数据平台。目前已有10种食用菌完成了基因组测序,虽然有多个实验室在开展基因敲除、RNA干扰研究,但由于食用菌的基因被敲除和干扰的效率低,这么多年了食用菌功能基因组研究未见突破性进展。本研究拟采用2013年初建立起来的第三代人工核酸内切酶CRISPR/Cas9系统来建立食用菌基因功能鉴定技术平台,为食用菌的功能基因组研究提供技术与方法。

英文摘要: Genome sequencing technology has become increasingly mature, and the cost has been greatly reduced. Genome sequencing will become a basic data platform of molecular biology research in edible fungi. Ten genomes of edible fungi have been completely sequenced. Although several laboratories have devoted themselves to the research on gene knockout and RNAi, there was not big breakout in the research of funtional genomics of edble fungi for their poor efficiency .In this proposal,we aim to create a technology platform for functional gene identification of mushroom using the third generational endonucleases system - CRISPR/Cas9 which was built in 2013. This system is also expected to provide technology and method for functional genomics research of edible fungi.
