中文摘要: 作为严重的社会问题和医学问题,酒依赖的发病机制尚不明了。本课题拟应用遗传学、磁共振波谱技术和行为医学方法,研究中国汉族酒依赖患者代谢型谷氨酸受体3单核苷酸多态性对酒依赖戒酒前后不同时期磁共振波谱和认知功能的影响;酒依赖患者戒酒前后不同时期磁共振波谱变化与认知功能改变的关系。从生物学机制上探讨基因是如何影响酒依赖的发生和发展的规律,探讨酒依赖患者脑神经代谢的特点,以及酒依赖戒断后脑生理功能的恢复情况。本课题率先将遗传学、功能影像学和行为医学三者结合,开创了全新的研究领域,为酒依赖的病因学研究提供了新的思路和研究方法。研究将筛选出新的酒依赖易感基因位点,为评价患者的发病和复发风险,探讨基因治疗方法,开发新型戒酒药物奠定基础。

英文摘要: As a serious social and medical problem, the mechanism of alcohol dependence is still unknown. The objective of this project is to study the brain function changes used by Magnetic Resonance Spectroscopy (MRS), cognitive assessment and single nucleotide polymorphisms (SNPs) in the metabotropic glutamate receptor 3 (GRM3) genes, the relationship between MRS changes and cognitive functions for the different stages of abstinence in alcoholic patients, using the combined methods of genetics, MRS and behavioral medicine. The study is to explore how the genes affect the produce and development of alcohol dependence, the characteristics of nervous metabolism, the physiological recovery after abstinence from alcoholism. It’s a new research field which combines the genetics, MRS and behavioral medicine. This study will produce new way of thinking and research method. This study will find new susceptibility gene locus for assessment of risks of development and relapse of alcohol dependence, future genetic therapy and the produce of new anti-alcoholic drugs.
