中文摘要: 本课题在前期证实透骨消痛胶囊能推动软骨细胞周期的进程、维持软骨细胞表型稳定、有效调控软骨细胞凋亡的基础上,针对软骨细胞生成和分化中有重要影响的Wnt/β-catenin信号通路进行探讨。课题采用体内外实验观察透骨消痛胶囊及其拆方对SD大鼠膝骨性关节炎软骨细胞Wnt/β-catenin信号传导通路中相关基因的影响,探讨透骨消痛胶囊是否对Wnt/β-catenin信号转导通路具有调控作用,探明其保护关节软骨的作用环节;借助生物网络技术,构建透骨消痛胶囊及其拆方和Wnt/β-catenin信号传导通路的关系网络,并通过计算打断上述网络中各中药节点的网络效率,研究复方中存在的协同作用,为中医药防治骨性关节炎提供一个新依据。

英文摘要: In our previous studies,the results showed Tougu Xiaotong Capsule(TGXTC) could advance the process of chondrocytes,maintain their stable phenotypes and effectively regulate the chondrocyte apoptosis. Wnt/β-catenin signaling regulates proliferation and differentiation of chondrocytes.In this work,we evaluate the effects of TGXTC and its separate components on expression of related genes in Wnt/β-catenin signaling pathway in osteoarthritis chondrocytes of SD rats in vitro and in vivo,which explore if TGXTC can effectively protect articular cartilage through regulating Wnt/β-catenin signaling pathway and the reasons of protective effect on articular cartilage. The interactions among TGXTC,its separate components and Wnt/β-catenin signaling pathway are mapped into a network by the technology of biological network. Furthermore,the synergistic effect in TGXTC is investigated by network efficiency calculation.Above work will provide a new basis for treatment osteoarthritis with traditional Chinese medicine.
