中文摘要: 猪繁殖与呼吸综合征病毒(PRRSV)的抗体依赖增强作用和抗原易变特性给PRRSV的疫苗研究和疫病控制带来了重重困难,给养猪业造成的损失巨大。大量研究表明,细胞免疫可能在PRRSV免疫保护中起到重要的作用,PRRSV所有的结构蛋白基本上都能诱发机体的细胞免疫反应。迄今为止,有关GP2a、GP2b、GP3和GP5a的T细胞表位还未见报道,而已报道的预测法鉴定的GP4和N蛋白的T细胞表位可能存在疏漏。因此,本项目拟通过重叠多肽法和IFN-γ酶联免疫斑点试验(ELISPOT)筛选法鉴定PRRSV GP2a、GP2b、GP3、GP4、GP5a和N蛋白的T细胞表位,并结合已报道的用重叠多肽法鉴定的GP5和M蛋白的T细胞表位,绘制PRRSV 结构蛋白的T细胞表位图谱。项目的实施将为研究PRRSV细胞免疫机制打下基础,对进一步研发PRRSV通用新型疫苗,有效防控PRRSV具有重要意义。

英文摘要: The charactitics that porcine reproductive and respiratory syndrome virus (PRRSV) had the antibody-dependent enhancement and the antigens of PRRSV were easily variable made it difficult to explore the effective PRRSV vaccine and to control PRRSV,so PRRSV made the huge economic loss for the swine industry.Many previous studies have shown that the cellular immune response may play an immportant role in controlling PRRSV, and most of all PRRSV structural proteins could induce the cellular immune response. However, as so far, there is no report about the T cell epitopes of PRRSV GP2a、GP2b、GP3 and GP5a, and the prediction method by previous report may miss some T cell epitopes of GP4 and N proteins. So the aims of present project were to screen and to identify T cell epitopes of PRRSV GP2a、GP2b、GP3、GP4、GP5a和N proteins by overlapping peptide methods and the IFN-γ ELISPOT assay,and then to draw the T cell epitope mapping of PRRSV structural proteins by adding additonal T cell epitopes of GP5 and M which have been reported. The implementation of the project will afford basis for studying the mechanism of cellular immunity of PRRSV and will be significant for futher exploring universal and noval PRRSV vaccine and for effectively controlling and preventing PRRSV.
