中文摘要: 脑卒中存在着发病率高、致残率高和死亡率高的现象,是继癌症和心血管疾病之后造成人类死亡和残疾的第三大原因。其中缺血性脑卒中占80%。为了更深入地阐明缺血性脑卒中的病理生理机制,本项目拟采用MALDI质谱成像这种高灵敏度和高通量的技术,从蛋白质水平上快速准确地对缺血性脑卒中进行研究。首先我们建立大鼠中动脉栓塞模型,通过冲洗预处理去除盐类,脂类等影响蛋白质质谱信号的化合物分子。然后采用蛋白酶对组织中的蛋白质进行原位水解,得到其肽段分子。最后我们将处理后的组织切片进行MALDI质谱成像,将肽段的质谱数据和蛋白质数据库相结合,我们可以得到与缺血性脑卒中相关的蛋白质的变化和空间分布信息。由于本项目研究操作简便,快速,能快速准确的发现与缺血性脑卒中相关的蛋白质分子,如获成功,有望代替传统的分析技术和生物手段用于缺血性脑卒中病理生理研究。

英文摘要: Stroke ranks only behind cancer and cardiac disease as one of the leading causes of death and a major cause of disability, with high incidence, morbidity and mortality. Matrix-assisted laser desorption/ionization (MALDI) mass spectrometry imaging (MSI) will be applied in the present study for investigation of the pathophysiological mechanisms underlying cerebral ischemia from protein levels. Brain sections form the middle cerebral artery occlusion (MCAO) rat model will be washed to remove salts, lipids, and other interferences, following by on-tissue digestion with enzymes. Peptides will be analyzed using MALDI MSI and proteins that are involved in pathophysiological processes of cerebral ischemia will be identified by searching for protein databases. Since the operation is simple, proteins involved in pathophysiological processes of cerebral ischemia can be rapidly identified. If this research is successful, it has promising application for investigation in pathophysiological mechanisms of cerebral ischemia in place of traditional analytical and biological technologies.
