中文摘要: 肿瘤标志物的多靶标灵敏检测是癌症早发现和早治疗的前提和基础,因此构建肿瘤标志物的多靶标免疫传感器,具有重大社会需求。本课题将分别以不同的氧化还原电对物质和不同金属/金属化合物纳米颗粒为电化学信号物质,对多种肿瘤标志物抗体进行编码,构建制备过程简便、灵敏、非酶、稳定的多靶标肿瘤标志物电化学免疫传感界面。利用中空纳米金负载氧化还原电对物质或者利用惰性纳米颗粒表面富集含不同金属/金属化合物纳米颗粒,并标记不同抗体,即一种氧化还原电对物质或一种含金属/金属化合物的纳米颗粒对应一种抗体,实现不同肿瘤标志物抗体的编码,研究构建肿瘤标志物多靶标免疫传感界面的新方法。系统研究电极表面的功能化设计,通过免疫识别性能与表面功能化的相关性,通过对传感界面的性能测试获得的信息,为优化多靶标电化学免疫传感界面的设计提供理论基础。

英文摘要: Sensitive detection of multi-target tumor markers is the prerequisite and basis for the early diagnosis and treatment of cance. Construction of electrochemical immunosensor has significant needs of the community. The different redox pairs of substances or metal/metal compound nanoparticles will be used as electrochemical signal substances to encode multiple antibodies of tumor markers, constructing electrochemical immunosensing interface for multiple tumor markers, which is simple, sensitive, non-enzymatic, and stable. To encode different tumor marker antibodies, this project will make use of hollow nanogold to load redox pairs substances and mark different antibodies or use of inert nanoparticles surface covered with different metal/metal compound nanoparticles marked different antibodies. That is, a redox pair substance or a metal/metal compound nanoparticles corresponds to an antibody. New method constructing immunosensing interface for multiple tumor markers will be investigated. A theoretical basis for optimizing the design of multi-target electrochemical immunosensor will be provided through systematic study on the functional assembly of the electrode surface, the correlation of immunol recognition properties and surface functional performance, and the information obtained by the performance testing of the
