中文摘要: 脑心肌炎病毒(EMCV)是近十年来我国猪群中新出现的又一种重要病原,能引起宿主I型干扰素(IFN)下调,从而抑制机体先天性免疫应答。前导蛋白L是EMCV重要蛋白之一,在病毒抵抗宿主防御系统中发挥着重要作用,但其在先天性免疫应答方面的作用尚无报道。我们采用Luciferase试验发现,EMCV L蛋白能够影响IFN-β启动子活性。本项目拟将进一步研究L蛋白引起机体I 型 IFN信号通路下调的分子机制;同时,从I 型 IFN发挥效应的信号通路上,尤其是JAK-STAT 信号通路来探索L蛋白如何影响I 型 IFN发挥效应,系统阐明EMCV的L蛋白影响机体I型 IFN信号通路的分子机理,丰富EMCV致病机制的理论基础。

英文摘要: Encephalomycoarditis virus (EMCV) is a new important pathogen of pigs in China. The virus infection could down-regulate type I interferon (IFN) to disarm host innate immune response. The EMCV leader protein L play important role in resist in host defense systems. But its foundation in regulating native immunity has not been reported. Recently, we found that the activity of IFN-β promoter could be decreased siginfacntly in BHK21 cells transfected with plasmid expressing L protein by using luciferase assay. This proposal will study on the molecular mechanisms of EMCV L protein down-regulating type I interferon by studing associated signaling pathways and IFN-mediated JAK-STAT signaling pathway. It should be usfull to elucidate the mechanism of EMCV L protein to antagonize the type I IFN signaling pathway and evade the host innate immune system, which will provide new data about the pathogensis mechanism of EMCV in the future.
