中文摘要: 胰岛素抵抗在Ⅱ型糖尿病的发生发展过程中起关键作用。蛋白酪氨酸磷酸酯酶1B(PTP1B)是胰岛素作用受体通路的主要负性调控因子,已成为Ⅱ型糖尿病药物研究的重要靶标。本课题组在前期研究中,从木豆叶中获得了一系列结构新颖的二苯乙烯类化合物,部分化合物具有显著的降血糖活性;初步的作用机制研究表明,此类化合物可通过促进胰岛素信号转导、抑制PTP1B活性而发挥降血糖作用。本项目拟在前期工作基础上,进一步对木豆叶中的二苯乙烯类化合物进行系统分离,并评价和研究该类成分抑制PTP1B的生物活性、作用机制及构效关系,为研究和开发选择性高、毒副作用小的PTP1B 抑制剂类抗糖尿病药物提供科学依据。

英文摘要: Insulin resistance plays a key role in the development of type Ⅱ diabetes. PTP1B (protein tyrosine phosphatase 1B) mainly take part in the insulin-dependent signaling cascade and are considered as a possible target for the treatment of type 2 diabetes. In our previous study, we have found a series of stilbenes from the leaves of Cajanus cajan with potent hypoglycemic activity. The preliminary study on the action mechanism indicated these stilbenes can inhibite the activity of PTP1B. Based on the previous study, this project will further isolate and identify stilbenes from the leaves of Cajanus cajan as well as evaluate their PTP1B inhibitory activities. Besides, we will also focus on analysis of their structure-activity relationship and elucidation of the molecular mechanisms underlying the regulation of PTP1B. Our study will provide the scientific basis for the development of novel antidiabetic lead compound with clear targets and distinct mechanisms.
