中文摘要: 膝关节骨性关节炎(Knee Osteoarthritis, KOA)是运动系统的常见多发病,女 性患病率高于男性,女性绝经后的患病率更明显高于同龄男性。本项目拟针对男女 KOA 患病率的差异,采用汉族背景的男女性不同影像学分级 KOA 的关节滑液为研究对象,以正常膝关节滑液为对照,运用定量蛋白质组学 iTRAQ 技术构建绝经前、后女性和同龄男性在 KOA 发生发展过程中的差异蛋白表达谱,筛选并验证由于性别差异所导致的与 KOA 发生发展相关的差异蛋白分子。结合血清性激素及关节滑液受体水平的测定,通过生物信息学方法解析差异表达蛋白分子与性激素水平相互联系及其可能的调控网络。本项目以性激素及其受体水平与KOA 发生发展的互作用为切入点,旨在揭示男女人群 KOA 发生发展过程中的差异蛋白分子基础及可能的调控网络,为 KOA 的发病机制研究提供新的理论依据和实验基础。

英文摘要: Knee osteoarthritis (KOA) is a common and frequently musculoskeletal disease among the elderly. Women had generally higher prevalence rates than men, especially after menopause. This study aimed toexplore the mechanisms underlying hormone and KOA by using ITRAQ quantitative proteomics technology. The subjects were the synovial fluid of different imaging of KOA among Han Chinese men and women, and the controls were the synovial fluid collected from healthy volunteers. Protein expression profiles during KOA occurrence and development between the women and men of the same age were analyzed by using ITRAQ quantitative proteomics technology, and the different protein molecules lead to the KOA were screened and certificated. Both the synovial fluid andthe serum sex hormone receptors were measured, and their possible regulatory network of the different protein molecules expression levels and serum sex hormone levels was analyzed using the bioinformatic analysis. The starting point of this study was the interaction the hormone and its receptor levels with KOA, and this study was designed to reveal the differences in the molecular basis of protein and possible regulatory network during KOA occurrence and development between the male and female. These data might contribute to increase our knowledge of the mechanisms underlying hormone and KOA, and also might provide some potential treatments to KOA.
