中文摘要: 如何在保证猪的生长速度和胴体瘦肉率的经济性状基础上,提高猪的肉质水平已成为21世纪育种专家新的攻关方向。猪肉质性状属于典型的数量性状,表观遗传状态是调控微效多基因表达的主要分子机制之一。哺乳动物的表观遗传模式主要在胚胎早期细胞分化过程中建立,并一直影响着性状基因的表达。叶酸作为S-腺苷甲硫氨酸的甲基提供者,是影响表观遗传状态主要营养成分之一,妊娠早期母猪的叶酸营养是否通过胎猪的表观遗传状态影响着猪肉质水平国内外尚未见报道。随着猪基因组图谱的逐步明晰,本课题利用高通量技术研究妊娠早期叶酸缺乏对猪肉质性状的影响,寻找叶酸影响肉质的关键基因,探讨其影响基因表达的表观遗传学调控机制,目的在于阐明猪肉质性状改良的营养表观遗传学依据,为猪肉质性状改良提供遗传学和表观遗传学分子基础。本课题创新点:首次探讨母猪妊娠早期叶酸营养对改善肉质性状的重要性;首次在体外建立不同浓度叶酸影响骨骼肌细胞增殖分化的模型。

英文摘要: How to improve meat quality level, based on the economic characters of fast growth and high carcass lean meat, has become a new research direction for pig breeding experts in the 21st century. Meat quality is a typical quantitative traits and the epigenetic state is one of the major molecular regulation mechanism of gene expression. The mammalian epigenetic model is established in the cell differentiation process in early embryo, and has been affecting the expression of the trait loci in the whole life. As the methyl donor of SAM, folic acid is one of the most important nutrient ingredients affecting the epigenetic stat,however the effect of folic acid on meat quality traits and its epigenetic mechanism has not been reported at home and abroad. With the pig genome gradually clear, the using high throughput technologies, this project is to study the effects of folic deficiency during early pregnancy on the pork quality traits, to find the key genes of the effect of folic acid on the meat,and to explore epigenetic regulation mechanism of the gene expression related meat quality.
