中文摘要: 骨重建是由破骨细胞和间充质干细胞(MSC)来源的成骨细胞精确协调完成的。在破骨细胞骨吸收过程中,各种基质细胞因子从骨基质中激活并释放,从而诱导骨髓中的MSC迁移到骨吸收部位,参与骨重建。然而,MSC被动员前,其赖以生存并完成自我更新的微环境- - -"干细胞巢"是什么?MSC是如何被动员并脱离这个微环境的机制尚不明确。我们研究发现:骨髓腔中,MSC与血管内皮前体细胞(EPC)存在着高度黏附现象,而且MSC生物学行为相对稳定。当这种黏附行为解除后,MSC的某些生物学行为随之改变。因此假设:EPC可能是MSC"干细胞巢"的重要组成,是MSC赖以生存并完成自我更新的重要微环境。围绕此假设,本研究拟探讨MSC和EPC间的相互黏附作用及其分子机制,明确这种黏附对MSC的生物学行为的影响,并解释MSC被动员并脱离这种黏附的分子机制。从而,补充对MSC生物学行为的认知。

英文摘要: Bone marrow stroma is composed primarily of non-hematopoietic stromal cells (BMSCs), a subset of which is multipotent, able to differentiate into osteoblasts, chondrocytes, stromal cells that support hematopoiesis, and marrow adipocytes. The bone formation is achieved by BMSCs that are recruited to the bone resorptive sites by the release of factor(s) during osteoclastic bone resorption. However, it is still unclear yet what is the stem cell niche, the microenvironment in which MSCs locate and commit their self-renewal? And how are MSCs recruited out of the microenvironment. Previously, we found that there was a high interaction between MSCs and endothelial progenitor cells (EPCs) in the bone marrow microenvironment, and the biological behavior of MSCs were relatively stable. As the adhesive effect was broken, some of the biological behavior of the MSCs changed. Therefore, it is assumed that the EPCs may be an important component of the MSC niche, in which MSCs home and complete the self-renewal. Accordingly, this study is to investigate the interaction between the MSCs and EPCs and its molecular mechanism, to clear the role of the interaction in the biological behavior of the MSCs, and explain the molecular mechanism that the MSCs are recruited out of the niche.
