中文摘要: 随着纳米科技的迅速发展,无机纳米药物载体在药物输送系统中展现出巨大的应用前景,其生物学效应受到广泛关注。研究纳米药物载体与血浆蛋白的相互作用,有助于理解其在生物体内转运、分配、毒理及代谢过程情况。本课题选取具有代表性的无机纳米药物载体,从载体与血浆蛋白之间的界面组装能否改变蛋白应有的功能出发,将纳米化学与蛋白质化学相结合,研究无机纳米载体影响血浆蛋白的结构及功能的机理,建立无机纳米药物载体与血浆蛋白结合和载体影响蛋白功能的关系,明确无机纳米药物载体在血浆蛋白上的结合位点,从分子水平上直观的反映出不同类型的无机纳米药物载体的生物效应的差异,阐明该类纳米材料的化学生物过程机制,对无机纳米药物载体毒理、转导机制及长期的安全性评估具有重要意义,有利于未来新型生物医用材料和多功能药物载体的开发和研究。

英文摘要: With the rapid development of nanotechnology, inorganic nanocarriers in drug delivery system show great prospects and their biological effects should be highly concerned.The studies on the interaction of nano-drug carriers with plasma proteins can contribute to the understanding of the carrier's transporting, distribution, toxicity, and metabolic process in vivo.In this research, some inorganic nano-drug carriers will be selected to study the effects of the interface assembly between the carrier and plasma proteins on the biological functions of proteins.The combination of the nano-chemistry and protein biochemistry, studyies of the interaction mechanism of inorganic nano-carrier with plasma protein, establishment of the relationship between the binding interaction of inorganic nano-drug carriers with plasma proteins and the effects of the binding interaction on carrier protein function, explanation about the binding sites of the inorganic nano-drug carriers on the plasma proteins, and reflection on the biological differences of different types of inorganic nano-drug carriers from the molecular level can clarify the chemical and biological mechanism of inorganic nano-drug carriers.These studies on the toxicology of inorganic nano-drug carriers, the transduction mechanism and long-term security are of great sign
