中文摘要: 二维过渡金属二硫化物纳米片及其复合材料具有类石墨的独特的物理结构而具有优良的光学、电学、催化等性能,近来备受关注。本项目拟合成多种新型二维过渡金属二硫化物纳米片及其复合材料,利用抗体、核酸适体等对靶标蛋白质的特异性识别,结合多种信号放大技术(如底物催化放大技术、核酸末端保护分析技术、核酸内切酶辅助目标循环放大技术、滚环扩增技术、杂交链反应放大技术等),构建一系列简单、快速、高灵敏、高特异性的电化学生物传感新平台,实现对蛋白质的灵敏、通用性检测,并应用于肿瘤细胞、血样等实际样品分析,期望为癌症等重大疾病的早期诊断、临床治疗和药物筛选等提供简便实用的新方法和新技术。

英文摘要: Recently, two-dimensional transition-metal dichalcogenides nanosheets and its nanocomposites have attracted increasing attention due to excellent optical, electrical and catalytic performances that come from its unique physical structure. This project aims to synthesis of some novel two-dimensional transition-metal dichalcogenides nanosheets and its nanocomposites. A series of simple, rapid, ultrasensitive and universal electrochemical biosensing platforms will be developed for protein detection based on the new materials by specific recognition of antibody and aptamer combined with signal amplification strategies (such as the substrate catalytic technology, DNA terminal protection assay, nicking endonuclease-assisted target recycling amplification, rolling circle amplification and hybridization chain reaction amplification), and would be applied to real samples (cancer cell, blood, ect.) analysis. It is expected to provide simple and practical new methods and technologies for serious early diagnosis, clinical treatment and drug screening of derious diseases (such as cancer).
