中文摘要: 青藏高原野生大麦是我国独有的珍稀种质资源,已探明具有广泛的抗逆遗传变异,发掘其耐盐资源可为栽培大麦耐盐性改良提供优异的遗传资源。在前期对近200份青藏高原一年生野生大麦进行遗传多态性分析和耐盐鉴定,以及采用关联分析鉴定耐盐种质的基础上,本项目拟采用组学方法,分析和比较野生和栽培大麦在蛋白质组、离子组和代谢组水平上对盐胁迫的响应差异,主要研究内容有:(1)利用同位素标记相对和绝对定量(iTRAQ)技术,研究蛋白质组的响应差异,鉴定耐盐相关蛋白;(2)利用电感耦合等离子体发射光谱仪(ICP-OES),研究离子组的响应差异和盐胁迫下离子的转运和积累;(3)利用气相色谱-质谱联用(GC-MS)技术,研究代谢组的响应差异和盐胁迫对相关代谢途径的影响。本研究将阐明盐胁迫过程中大麦组织基因-蛋白-代谢物的分子响应,从而揭示野生大麦特异的耐盐机制,丰富植物耐盐的分子机理理论,为大麦耐盐性改良提供理论指导。

英文摘要: Tibetan wild barley (Hordeum vulgare ssp. spontaneum) is rare and unique wild germplasm in China, and it shows a wide genetic variation in the tolerance to harsh environments. It is possible for us to explore the elite germplasm in terms of salt tolerance from the wild barley. In our previous studies, an association analysis was used to identify and explore salt-tolerant germplasm based on the evaluation of salt tolerance among around 200 Tibetan wild barley accessions. Here, multiple plant 'omics' methods will be used to analyze genotypic difference in response to salt tolerance between elite wild barley and cultivated barley. The main contents in the study are as fellows: (1) using isobaric tags for relative and absolute quantitation (iTRAQ) technique to compare proteomic difference and to identify differential expressed proteins; (2) using inductively coupled plasma-optical emission spectrometer (ICP-OES) technique to compare ionomic difference and to investigate ion transporting under salt stress; (3) using gas chromatography-mass spectroscopy (GC-MS) technique to compare metabolomic difference and to reveal the impacts of metabolic pathway caused by salt stress.
