中文摘要: 受遗传-环境因素影响的青少年抑郁病因及发病机制不明确。研究表明早期应激与抑郁有关,5-HTTLPR\BDNF、CRHR、GR、FKBP5基因多态性各被发现与抑郁相关,结果多不一致,多基因研究甚少,但提示抑郁的遗传易感性。心理复原力可能有助于个体应对应激,正被广泛关注。早期应激与遗传易感性及心理复原力如何影响抑郁发生?其神经生物学机制是什么?研究甚少,特别对中国汉族青少年的相关研究尚未见报道。因此本研究在配比四组早期应激有抑郁、无早期应激有抑郁、无早期应激无抑郁、有早期应激无抑郁的青少年横断面分析以上相关因素与抑郁的基础上,对后两组进行2年半队列研究,分析早期应激、遗传易感性及心理复原力对抑郁的影响, MRI及fMRI研究分析脑情绪调节环路结构及功能的变化,依皮质醇水平评价HPA轴功能,探索早期应激与遗传易感性及心理复原力影响抑郁发生的神经生物学机制,以对青少年抑郁的预防及干预提供新的思路。

英文摘要: The pathogenesis and mechanism of depression for adolecent are still unclear. Some researches showed that depression was related to early life stress. Respective study results disagreed and contradicted on the relation of 5-HTTLPR\BDNF or CRHR or GR or FKBP5 genetic polymorphisms and depression involving in two systems of stress reactivity.Research on multiple genes related to depression is very few.However it suggests the genetic vulnerability of depression.Mental resilience contributing to cope with depression is being paid attention. How do early life stress and genetic vulnerability and mental resilience affect depression for adolescent? What is its mechanism of neurobiology? Researches on these aspects are few. Especially the related research for Chinese Han adolescent isn't seen to report at present. So in our study the four matched groups including early life stress-depression, no early life stress-depression, early life stress-no depression,no early life stress-no depression will be analyzed on above factors related to depression in the cross-sectional study.Then the later two groups will be used in the cohort study with 2.5 years. The purposes of our study are to understand the interaction of early life stress, genetic vulnerability and mental resilience on depression, the transformation of structure and function of brain emotional regulation circuit by MRI and fMRI study,the change of HPA function assessed by saliva cortisol,and to explore the neurobiology mechanism of the influence of early life stress and genetic vulnerability and mental resilience on depression.It will provide a new way of prevention and intervention for adolescent depression.
