中文摘要: 目的:通过观察清热解毒代表药物黄芩、板蓝根、白头翁、虎杖和白花蛇舌草对流感病毒FM1感染小鼠不同时相细胞炎性因子、抗炎因子动态变化的水平以及对肺、气管和支气管等器管组织病理损伤的影响,探讨清热解毒法对炎性细胞因子干预及免疫损伤保护的作用机制。方法:建立流感病毒亚甲型鼠肺适应株FM1感染BALB/cAnN小鼠模型,采用ELISA法、RT-PCR法、免疫组化法检测促炎因子TNF-α12289;IL-1、IL-6及抑炎因子IL-10、IFN-γ30340;动态变化,同时采用光学显微镜及电子显微镜下观察不同时相清热解毒代表药物对免疫损伤的修复作用。结果:流感病毒感染小鼠后,感染模型组的炎性因子在感染后第3、5天达到峰值,肺、支气管等组织器官病变严重,证实了"在热病热毒炽盛时期,也正是炎性因子异常增高,免疫反应激烈的时期"的假说。黄芩和板蓝根在流感病毒感染第3、5天降低可促炎因子,升高抑炎因子的水平;白头翁、虎杖和白花蛇舌草调节流感病毒引起免疫损伤的作用,不如黄芩和板蓝根。结论:清热解毒代表药物黄芩、板蓝根在流感病毒感染后第3天和第5天可明显下调促炎性因子并上调抗炎因子水平,从而较好的干预流感病毒引起的免疫损伤。

英文摘要: Objective: This project is to observe the effect of Scutellaria, Isatidis, Pulsatilla, Polygonum cuspidatum, Oldenlandia of removing toxic heat drugs on inflammatory cytokines , anti-inflammatory cytokines's level and lung,trachea , bronchus ect.impaired organ which infected by FM1 mice at different stage in order to disscuss the intervention mechanism of removing toxic heat method.Method: To establish FM1 infected mice model,use ELISA,RT-PCR,Immunohistochemical method to test flammatory cytokines and anti-flammatory cytokines,meanwhile,to observe the repair immune injury role of removing toxic heat drugs.After infected by FM1 mice,the FM1 infected group's flammatory cytokines was significantly increased when the mice was infected 3 or 5 days.Lung,trachea , bronchus was badly impaired.This result confirmed the hypothesis that "the stage of excessive heat and toxin was the time when cytokines were abnormally increasing and immune response was intense". Scutellaria and Isatidis decrease flammatory cytokines while increase anti-flammatory cytokines level when mice was infected 3 or 5 days. The effect of repairing immune injury of Pulsatilla, Polygonum cuspidatum and Oldenlandia was not as well as Scutellaria and Isatidis.Conclusion: Scutellaria and Isatidis of removing toxic heat drugs can significantly decrease flammatory cytokines while increase anti-flammatory cytokines level when mice was infected 3 or 5 days in order to repair immune injury.
