中文摘要: 制备兼具刺激响应药物释放和骨修复作用的多功能纳米结构给药系统是实现长效间接控制的药物治疗和替代性骨修复治疗协同作用的关键技术,其中生物安全性是须要优先解决的问题。本项目拟选择具有良好生物相容性和生物可降解性的层状硅酸钙作为主体材料、功能纳米粒子和生物高分子作为协助材料,以功能化的药物控释方案进行设计,通过功能纳米粒子表面嫁接、药物分子插层装载和生物高分子表面包覆,制备具有光/磁响应药物控释功能的硅酸钙基纳米结构夹层给药系统,解决该给药系统的关键制备技术,并阐明其中硅酸钙主体与功能纳米粒子、药物分子、生物高分子等客体之间的相互作用机制,以及功能纳米粒子的功能化刺激对药物释放的控制机制,为形成用于替代性骨修复治疗、具有我国自主知识产权的新型药物制剂与发展医疗新技术提供科学依据。

英文摘要: The preparation of multi-functional drug delivery systems combining stimuli-responsive controlled-drug-release and bone repair ability is the key technology to achieve the combination of long-term drug therapy and in situ bone repair. The biological security of such systems should be taken into account first. In this research, layered calcium silicate with good biocompatibility and biodegradability is to be utilized as the host material, and functional nanoparticles and biopolymers are to be employed as the assistant mateirals. For the purpose of stimuli-responsive controlled-drug-release, the drug delivery system that can be activated by infrared light or alternating magnetic field is to be prepared via three steps: a) the linkage of the functional nanoparticles to the surface of the host material, b) the intercalation of drug molecules into the layered nanostructure of the host material and c) the coating of the biopolymers onto the surface of the host material. The key technology for the preparation of such a drug delivery system is to be studied and the interaction mechanisms between the host material, assistant materials and drug molecule are to be demonstrated.
