中文摘要: 本项目依据氧化锰(MnO)粒子独特的T1磁学变亮成像模式和金(Au)粒子特有的表面等离子体共振(SPR)光学性质,采用高温热解法在有机相中合成具有"双向"结构的Au-MnO磁光双功能纳米颗粒;利用生物相容性聚乙二醇衍生物作为表面改性剂赋予"双向"Au-MnO磁光纳米颗粒生物相容性的表面,实现主动靶向性给药、高药物负载率及药物控制释放的目标。通过考察纳米粒子表面基团与抗癌药物不同的结合方式对药物控释效果的影响,探寻最佳的药物负载方式以降低药物剂量从而减轻毒副作用。以Sk-Br-3乳腺癌细胞和肿瘤小鼠为研究模型,利用激光共聚焦显微成像和核磁共振成像技术,研究"双向"Au-MnO粒子在癌症影像诊断中的应用。本课题提出的集T1磁学成像、光学成像和药物靶向控制释放于一体的新型纳米药物将为深化癌症的早期准确诊断及治疗提供理论基础和技术支持。

英文摘要: On the basis of the T1 unique magnetic bright imaging mode of manganese oxide (MnO) nanoparticles and the Surface Plasmon Resonance(SPR) optical properties of gold (Au) nanoparticles, we are devoted to synthesize "bi-direction" structure Au-MnO nanoparticles through a synthetic approach of pyrolysis. Then we incorporate biocompatible PEGylated derivatives as surface modifier to modify Au-MnO nanoparticles, thus giving the nanoparticles biocompatible surface, which can realize the active targeting drug delivery, high drug loading rate and controlled release of drugs. Through the investigation of the nanoparticles surface groups and anticancer drugs of different binding modes for drug controlled release effect, the optimal drug loading methods will be found to reduce drug doses and the side effects. By means of Sk-Br-3 breast cancer cells and tumor mouse as models to study the "bi-direction" Au-MnO particles in cancer imaging diagnosis application will be explored using laser confocal microscopic imaging and magnetic resonance imaging techniques. This subject with T1 magnetic imaging, optical imaging and targeting drug controlled release in a new nanomedicine system will provide theoretical basis and technical support to accurate diagnosis and treatment for cancers.
