中文摘要: 大量研究证明,非编码RNA可介导基因表达的调控,而基因表达的一个关键步骤是基因的转录。RNA聚合酶Ⅱ转录细胞中所有mRNA和很多非编码RNA基因。目前仅发现四个非编码RNA调控RNA聚合酶Ⅱ介导的mRNA转录的起始或延伸,它们的作用机制又各不相同。鉴于非编码RNA作用的广泛性和多样性,我们将筛选和鉴定其它调控RNA聚合酶Ⅱ转录的非编码RNA。我们的前期实验中发现了一批新的与RNA聚合酶Ⅱ结合的非编码RNA,包括一些长非编码RNA(lncRNA)。我们将从中鉴定出调控基因转录的非编码RNA,并探讨它们作用的分子机理,以及这种调控对细胞的生物学功能的影响。我们的研究将有可能丰富对非编码RNA调控mRNA转录机制的认识,并为申请人将来进一步研究非编码RNA调控基因的表达打下基础。

英文摘要: Multiple lines of evidence have shown that noncoding RNAs (ncRNAs) can regulate gene expression, and one crucial step of gene expression is gene transcription. All of the eukaryotic protein-coding genes and some noncoding genes are transcribed by RNA polymerase Ⅱ. So far, there are only four noncoding RNAs that have been found to directly regulate transcription initiation or elongation mediated by RNA polymerase Ⅱ. Considering the diversity and versatility of noncoding RNA effects, we determine to uncover ncRNAs that directly regulate gene transcription mediated by RNA polymerase Ⅱ. In our preliminarily data, we found a variety of noncoding RNAs including some long noncoding RNAs (lncRNAs) that bind with RNA polymerase Ⅱ. We will investigate which of and how these noncoding RNAs control gene transcription by RNA polymerase Ⅱ, and explore further the corresponding regulatory effect on cell physiology. This project could aid our understanding on the function and functional mechanism of the regulation by noncoding RNAs on pol II transcription, and we hope the completion of this proposal will lay a foundation for our future exploration on the regulation of gene expression by ncRNAs.
