中文摘要: 活的非可培养状态(VBNC)的大肠杆菌O157:H7在条件合适时能复活,从而恢复致病性,形成潜在重大的食品安全隐患,已引起研究者高度重视,成为近年来的研究热点。然而学术界对其复活的分子机制并不清楚,导致对该菌的VBNC状态检测和控制处于技术空白。本项目研究该菌在VBNC状态下,通过诱导复活过程中,不同阶段 VBNC菌与正常菌蛋白表达谱的差异,对特征蛋白进行功能鉴定与分析后,定位到相关功能基因,通过基因敲除,进一步确认这些功能基因在VBNC状态复活中的地位与作用,探明重要关键基因的定位与功能,尤其是关键基因之间组成的基因表达传递调控链脉络,揭示"蛋白(谱)-基因(谱)-CDS(群)-功能"的内在关系。在以上研究工作的基础上,最终阐明VBNC状态大肠杆菌O157:H7复活的分子机制,为食品中VBNC状态病原菌的检测与控制打下重要的理论基础,对其它VBNC状态病原菌的研究有很好的借鉴作用。

英文摘要: In recent years, research on Viable but nonculturable state (VBNC)of Escherichia coli 0157:H7 has been highlighted since VBNC bacteria can be resuscitated from dormancy in appropriate conditions leading to food safety issues. However the detection and control technology of the VBNC bacteria is still immature mainly because the mechanism of the resurrection process remains unclear. This research intends to demonstrate the protein expression diversity between the VBNC and normal bacteria during the various phases of the resurrection process, and find out the functional genes by protein identify and functional analysis. The crucial genes will be further analyzed by "gene deletion" to verify the restoration function and the expression regulation relationships of these genes will also be demonstrated for deep understanding the relevance among "protein- gene- CDS- function". This research will finally improve the understanding of molecular mechanism behind phenotypic changes in culturability restoration of VBNC E. coli 0157:H7,which can become a good basis for the determination and control of VBNC bacteria in food.
