中文摘要: 新疆煤炭资源丰富,占全国预测储量的四成以上,正大力发展煤电、煤气化、煤液化及煤焦化四大产业链。而煤燃烧过程中所产生硫污染已对环境造成严重影响,高效脱硫技术对于新疆经济可持续发展意义重大。生物脱硫技术虽然潜力巨大,但由于脱硫菌功能单一、脱硫效率低的问题,工业化应用十分有限,寻找和研究新型、高效、适合在工业生产中采用的生物脱硫方法是目前煤脱硫的研究热点之一。针对当前生物脱硫研究的现状,本项目基于筛选到的可降解有机硫和无机硫的微生物,考察不同脱硫微生物的脱硫机理,组合噻吩类脱硫酶基因和黄铁矿类脱硫酶基因,并通过插入信号肽的编码序列基因,构建复合脱硫重组菌,高效表达分泌型重组脱硫酶;研究重组脱硫酶的脱硫活性、稳定性及动力学特征,考察重组脱硫酶对原煤脱硫效果,优化酶法脱硫的工艺参数,提高脱硫效率,为原煤生物脱硫技术的工业化应用奠定基础。

英文摘要: Sulfur dioxide from coal burning brings serious environmental pollution. The desulfurization technology is of great significance for the sustainable economic development in Xinjiang, abundant in coal resources. Microbial desulfurization technology has great potential in this area, but desulfurization of industrial application is very limited. New, efficient and suitable for biological desulfurization method used in industrial production is quite important. Screening microbial degradation of the organic sulfur of the thiophene and inorganic sulfide will be carried out and the mechanism of microbial desulfurization will be investigated. Different desulfurization genes are constructed inone engineering strain, and through the introduction of signal peptide coding sequence genes, the new composite recombinant strain will be constructed, efficient expression of the secreted recombinant desulfurases. Desulfurization activity and kinetic characterization of recombinant desulfurases will be investigated. Combined with the water washing coal slurry transportation, optimizing operation parameters will be achieved for the efficient raw coal desulfurization.
