中文摘要: lncRNAs是一类种间保守性相对较低的长链非编码RNA,作为生物体内重要的调节因子,可以通过多种途径参与机体的各类生命活动。在前期研究中,本实验室利用lncRNAs深度测序对牛不同生长发育阶段肌肉组织中的lncRNAs进行检测,获得多个差异表达lncRNAs。本项目拟在此基础上,利用生物信息学分析及q-PCR方法鉴定牛肌肉发生过程中的关键lncRNAs,通过构建牛成肌细胞lncRNAs过表达和功能缺失模型,验证lncRNAs对牛肌肉细胞增殖和分化的影响,并利用RNA pull-down 和质谱技术获取并辨别lncRNAs的互作蛋白,研究 lncRNAs与其靶标蛋白的作用模式,阐明其调控通路,系统分析牛肌肉发育过程中关键lncRNAs及其靶标蛋白功能。

英文摘要: LncRNAs are a class of lowly conserved, long non-coding RNAs that emerge as a key regulator involved in multiple cellular processes by various ways. Our previous study revealed many lncRNAs expressed differently among different developmental stages of bovine muscle by lncRNA-seq. In this study, we will identify the major lncRNAs in cattle myogenesis by bioinformatics analysis and q-PCR and then construct gain of function and dysfunctional cell model in order to investigate the effect of lncRNAs on the proliferation and differentiation of bovine myoblasts. Furthermore, RNA pull-down trail and mass-spectrometric analysis will be carried out to recognize lncRNA’s potential target protein and determine the relationship between them. Integration of all results, we will systematically elucidate the roles of lncRNAs and their target protein play in bovine muscle development.
