中文摘要: 环境中许多污染会对生物体中枢神经系统产生影响,其作用机制十分复杂,而神经递质极有可能在其中起着关键作用。多巴胺(dopamine, DA)是中枢神经系统关键神经递质之一。近期研究显示,多巴胺系统很可能是环境污染物(如双酚A、多氯联苯)作用的重要靶点之一。典型的环境污染物多溴联苯醚(PBDEs)是电子垃圾产生的最主要有机污染之一,在全球特别是我国的环境中长期存在,并且已被多次证实具有神经毒性。考虑到与多氯联苯的结构相似性,PBDEs也很可能对多巴胺系统产生影响。本课题拟采用斑马鱼为模式动物,通过环境浓度水平下的急性/长期暴露,研究PBDEs对斑马鱼多巴胺系统的发育和正常功能的影响,并寻找PBDEs作用的主要位点,同时证实多巴胺系统损伤是PBDEs暴露对斑马鱼产生神经毒性的重要途径之一,并探索可能的分子机制。

英文摘要: Various environmental pollutants could disrupt the central nervous system (CNS) in both animals and human beings, and the potential mechanisms can be very complicated, in which the neurotransmitters may play an important role. Recently, dopamine, one of the key neurotransmitter in the CNS, has been identified as target of environmental pollutants, such as bisphenol A and polychlorinated biphenyl (PCBs). Considering the similarities in physicochemical properties, polybrominated diphenyl ethers (PBDEs), one of the main neurotoxins widely found in e-waste recycling regions, might also have an effects on the dopamine system. For this purpose, we intend to use zebrafish as model animals, to investigate the effects on the development and biological functions of the dopamine system by PBDEs at environmentally relevant concentrations, and to verify the presumption that damages in dopamine system might contribute, at least partially, to the neurotoxicity by PBDEs exposure.
