中文摘要: 稀土上转换发光纳米材料的信噪比高、发光稳定性好、激发光源(近红外半导体激光器)价廉易得,具有重要的研究意义和应用潜力。然而,目前该材料多使用生物组织吸收较强的980 nm激发,限制其穿透深度和发光效率的提高,难以满足实际需求。本项目拟通过选择合适的基质,掺杂Nd3+和其他稀土离子,利用多层核壳结构及Mn2+过渡金属离子,制备出808 nm 激发下高强度红光发射的稀土上转换发光纳米材料。通过合适的表面修饰方法,实现该材料在小动物活体深层组织癌细胞的光动力学治疗及生物成像。本项目的开展能有效消除生物组织对近红外光的吸收造成的温度过高和穿透深度浅等缺陷,有利于实现癌细胞活体同步增强荧光成像和光动力学治疗,对扩展稀土上转换发光纳米材料在生物领域的应用范围具有一定的指导价值。

英文摘要: Lanthanide-doped upconverting nanomaterials have important significance and great potential applications owing to their high photostability, low-cost excitation source, and high signal-to-noise ratio. However, the frequently-used 980 nm excitation source largely overlaps the absorption band of water molecules that are dominant in biological samples, which would be significantly attenuated while passing through biological samples. This project aims at preparing high efficient red-Emission of lanthanide-doped upconverting nanoparticles under excitation at 808 nm by doping Nd3+ and multi- core-shell structure in suitable host materials to achieve photodynamic therapy and bio-imaging in deep-tissue cancer. The deep-tissue cancer photodynamic therapy and imaging based on the obtained nanomaterials suitable surface modification will be carried out. The development of this project may be benefit for simultaneously enhancement of deep-tissue fluorescence imaging and photodynamic therapy, and eliminates defects of overheating and penetration issues resulting from absorption of near-infrared light by biological samples. It also may be valuable for expanding the application in biological area based on lanthanide doped upconversion materials.
