中文摘要: 本课题用Caco-2单分子层模型进行花色苷的吸收、转运和首过代谢研究。建立花色苷黄烊盐化方法,获得准确的花色苷检测和鉴定方法;通过Caco-2体外模型和在体灌肠模型,探索花色苷结构对其吸收和转运影响;通过花青素慢性暴露处理及转运体特异阻断手段寻求花色苷特有的体内主动运输新转运体;利用HPLC-DAD-ESI-MS检测和鉴定花色苷在吸收转运过程中的首过代谢问题。项目的实施对阐明花色苷在小肠的吸收和转运的机制和揭示花色苷低生物利用度现象具有重要意义,为提高人们膳食中的花色苷生物利用度奠定基础,为人们在日常生活中能够更好地利用膳食中的花色苷而合理地设计膳食结构和加工方法提供思路。

英文摘要: The project will study the absorption and the transportation of anthocyanins by the Caco-2 monolayer model, and the first-pass metabolism of anthocyanins in Caco-2 cell will also be studied. The conditions will be found to transfer the anthocyanins to flavylium anion isoform for the accurate determination of anthocyanins. The affection of the structure anthocyanins on their absorption and transport will be explored by Caco-2 monolayer model and in situ rat single-pass intestinal perfusion model. Chronic exposure treatment and transporter -specific blocking means are used to find the new specific transporter of anthocyanins. The metabolism of anthocyanins in the Caco-2 during the transport will be determined by HPLC-DAD-ESI-MS. This project is important to clarify the intestinal absorption and transport mechanism and the low bioavailability of anthocyanins, which are rich in the food of people. The results will lay the foundation for improve the bioavailability of anthocyanins in people’s diet. And it will provide good ideas and processing methods to make better use of anthocyanins for people in their daily diet. The reasonably designed diet will improve the health of people.
