中文摘要: 厌氧菌硫酸盐还原菌(SRB)是导致金属腐蚀的主要微生物种群之一。对SBR形成生物膜并造成金属腐蚀的分子机制目前还很不清楚,限制了抗微生物腐蚀制剂的开发和应用。本申请计划使用SRB模式菌普通脱硫弧菌(Desulfovibrio vulgaris)为研究对象,研究细菌中广泛存在的信号转导系统-双组分调控系统(TCSs)对微生物金属腐蚀过程的调控机制,进而发现开发新的抗腐蚀制剂的基因目标。研究基于以下两个科学假设:一)TCSs 参与调控脱硫细菌生物膜生长及所引起的金属腐蚀;二)生物膜不同部位的细菌的基因表达存在异质性,不同部位的细菌对金属腐蚀的作用有所不同。本研究将结合突变株分析,转录组学以及单细胞分析等多种方法,发现和鉴定在D. vulgaris中与生物膜形成及金属腐蚀相关的TCSs及其下游相关基因,提高在遗传学水平上对普通脱硫弧菌生物膜形成的分子机制的理解,为金属腐蚀防护提供必需的理论基础。

英文摘要: The anaerobic sulfate-reducing bacteria (SRB) is one of the major microbial groups that cause iron corrosion. So far very little is known about the regulatory mechanisms governing biofilm formation and its corrosion activity on iron, which has slowed down various efforts of developing more efficient corrosion inhibitors. This proposal aims at deciphering regulatory mechanisms of microbial corrosion by two-component regulatory systems (TCSs), and identifying new corrosion control targets in the model SRB, Desulfovibrio vulgaris. The research is proposed based on two scientific hypotheses: i) TCSs proteins may be involved in regulation of biofilm formation in D. vulgaris and its corrosion activity on iron; ii) gene expression heterogeneity exists in different parts of biofilms and bacteria in different parts of biofilms play varying roles in iron corrosion. The proposed study will combine mutant analysis, transcriptomics and single-cell based gene expression profiling technologies to identify TCSs and their downstream genes related to biofilms formation and corrosion activity in D. vulgaris. The research will improve the understanding of the molecular mechanisms of SRB biofilm formation, and eventually provide most needed theoretical foundation for iron corrosion control.
