中文摘要: 干旱是影响农作物产量的非生物逆境之一。植物旱胁迫响应机制的研究集中在依赖ABA和不依赖ABA两条途径上。其中在依赖于ABA途径中bZIP转录因子可以通过调控下游基因的表达使植物在逆境胁迫中存活下来。已有的研究表明很多bZIP转录因子与旱胁迫相关,但对这些转录因子的下游靶基因及其调控机理的研究很少。本项目分别将转录激活序列VP16和转录抑制序列EAR与OsbZIP12融合,在玉米ubiquitin启动子的调控下进行超表达,转化水稻植株后,寻找转基因水稻中OsbZIP12的下游靶基因,分析旱胁迫相关生理指标,进一步揭示OsbZIP12在旱胁迫响应过程中的作用机理,为抗旱新品种的分子育种做出理论指导。

英文摘要: Drought is one of the major abiotic stresses that influences crop yield. Both ABA-dependent and ABA-independent processes are involved in stress responses. In the ABA signaling-mediated stress responses, bZIPs transcription factors have crucial regulator roles in activating ABA-dependent stress-responsive gene expression. In rice, many bZIPs have been proved to have functions in drought stress responses, but the target genes of these bZIPs are rarely found. We constructed the overexpression of OsbZIP12 fused with tetrameric VP16 activation domains (VP64) and EAR transcriptional repression domain respectively. After introducing the construct into rice, we will find the target genes of OsbZIP12, and focus on characterization of the physiological and molecular mechanisms under drought stress. Our work will provide theoretical guidance for manufacturing drought stress resistant crops in the field.
