中文摘要: 糖尿病引起的心血管疾病是导致患者致残、致死的主要原因之一。Ghrelin是一个具有促进生长激素分泌、增进食欲、促进脂类吸收和脂肪形成的小分子活性肽,与心血管的能量代谢和2型DM的胰岛素抵抗密切相关。我们的前期研究表明,Ghrelin能够抑制高糖高脂诱导的心肌细胞胰岛素抵抗,但其机制尚不清楚。本项目拟以体外培养心肌细胞为研究对象,应用形态学、生物化学和分子生物学等技术,研究Ghrelin对PI3K/AKT及其下游GLUT4、PPAR-γ、AMPK等通路蛋白的表达或活性的影响,阐明Ghrelin抑制糖脂毒性胰岛的心肌胰岛素抵抗的分子机制,为将Ghrelin作为预防和治疗DM心肌损害的新靶点提供理论基础。

英文摘要: Heart-blood disease induced by diabetes mellitus(DM) is one cause of disabality and death. Ghrelin is a micromolecule peptide which can improve growth hormone secretion and appetite and lipide absorption and lipid formation.It is related with heart-blood energy metabolism and insulin resistance of 2 type DM. We found ghrelin can inhibit insulin resistance induced by glucotoxicity and lipotoxicity in cardiomyocyte in early study. But the mechanism is not clear. The object of study is cardiomyocyte cultured in vitro. We utilize the methods of morphology and biochemistry and molecular biology to study the effect of ghrelin on PI3K/AKT pathway and the iter protein ofPI3-kinase/AKT pathway glucose transporter 4 (GLUT4), peroxisome proliferators(PPARr) and AMP activated protein kinase (AMPK) genes in downstream. The aim is to elucidate the molecule mechanism of ghrelin to inhibit insulin resistance of cardiomyocyte and to provide the rationale of the new target in prevent and treat myocardial damage of DM of ghrelin.
