中文摘要: 单增李斯特菌是威胁人类健康的重要食源性病原菌,死亡率高达20%以上,对食品中单增李斯特菌及其生物标志物的快速检测是保障食品安全的重要措施。本研究拟合成一个全长78bp中间含40个随机序列的随机单链寡核苷酸(ssDNA)文库,采用指数富集配体系统进化技术筛选单增李斯特菌的适配体,以Pb2+的竞争性剪切和硫酸二甲酯标记等方法探索适配体二级结构及与功能的关联性,为新型配体的获得提供技术借鉴。亲和素标记适配体作为捕获链,同链霉素亲和素标记的纳米磁性微球相连,同时合成侧翼与捕获链互补的ssDNA并标记荧光素cy3作为显色链,组装磁性微球适配体传感器。与不同浓度的单增李斯特菌发生反应,建立李斯特菌数量与荧光强度之间的关系,实现食品中单增李斯特菌快速和定量检测。

英文摘要: Listeria monocytogenes are important foodborne pathogens。The mortality is more than 20% .So the rapid detection of Listeria monocytogenes and its biomarkers in food is an important measure to protect food safety. This study would synthesis a random single-stranded oligonucleotide (ssDNA) library, which were 78base pairs and containing 40 random sequences.The exponential enrichment ligand phylogenetic screened aptamers. The secondary structure was determined by lead cleavage and dimethylsurfate modificationg.This provided a reference for screening new ligand.Aptamers were labeled with avidin as the capture chain, connected with the streptomycin-labeled magnetic beads. A complementary strand was labelled with cy3 as detecting signal. This was a aptamer sensor based on magnetic microspheres. The sensor reacted with different concentrations of Listeria monocytogenes, in order to establish the quantitative relationship between the hly concentration and fluorescence intensity of Listeria monocytogenes to achieve the purpose of rapid and quantitative detection of foodborne pathogens.
