中文摘要: 根直接影响植物从土壤里吸收营养,深入研究调控植物根部发育的分子网络,将为发展作物营养高效新农业提供重要理论基础。植物的根是由根分生组织细胞不断分裂和分化的结果,这个过程受到植物体的高度精细调节控制。目前在模式植物拟南芥中已经分离出一系列参与调控根发育的关键因子包括SCARECREW,SHORT ROOT(SHR),WOX5等,但其精细的调控机制有待进一步研究。申请人前期利用拟南芥关键因子SHR通过酵母双杂交筛库,获得了若干SHR结合蛋白 (SHR-Interacting Proteins, SIPs)。通过对其中一个互作蛋白SIP1的初步研究发现,该突变体的根分生组织发育受到严重影响,表明SIP1是控制根分生组织发育的关键基因。本项目将运用分子生物学、细胞生物学及生化等手段深入研究SIP1的生物学功能,揭示SIP1-SHR-SCR蛋白复合体在根分生组织发育过程中的调控作用。

英文摘要: Development of plant root directly impacts the absorption ability of nutrition from the soil. Understanding of the molecular regulatory mechanisms of root development will provide us an important theoretical basis for improving nutrient using efficiency of crops. At present, series of key factors including SCARECREW, SHORT ROOT (SHR), WOX5 and so on were reported to involved in root development in the model plant Arabidopsis thaliana. However the regulatory mechanisms are still largely unknown. Several interacting proteins (SHR-Interacting Proteins, SIPs) were isolated by yeast-two-hybrid screening. One of them, named SIP1, is a transcriptional factor of which biological function on root development is unknown. The root meristem of sip1 mutant shows severely affected, indicating that SIP1 is a key regulator of controlling root meristem development. In this project, we will study the biological function of SIP1-SHR-SCR protein complex by molecular biological, cell biological and biochemistry methods. This research will uncover the regulatory mechanism of root meristem development and would allow plant breeders to define the gene targets for the development of nutrient using efficient crops in future.
