中文摘要: 类风湿关节炎(RA)是临床主要致残性疾病,目前尚无特异性治疗方法。胶原性关节炎(CIA)为RA发病机制及治疗研究提供了理想的实验平台。自身反应性CD4+T细胞处于RA/CIA发病的中心环节,抑制其增殖及分化可能是从根本上特异性抑制RA免疫反应的有效途径。微小RNA(miRNA)是调控T细胞增殖和分化的重要分子,其在RA/CIA自身反应性T细胞中的意义还有待深入研究。本课题组预实验发现miR-144等与RA/CIA的T细胞调控相关,本课题进一步通过微阵列法分析CIA鼠脾CD4+T细胞的miRNA表达谱,T细胞功能实验验证miR-144等T细胞调控相关miRNA对CIA鼠CD4+T细胞增殖和分化的影响,并通过差异mRNA表达及生物信息学方法,分析miR-144等T细胞调控相关miRNA的作用机制。本课题不仅为miRNA在RA发病中的意义提供实验依据,而且为从起始环节特异性治疗RA提供新的思路。

英文摘要: Rheumatoid arthritis is a main disease which causes disability. There is still not specific therapy now. Collagen-induced arthritis (CIA) is a reasonable experimental arthritis animal model for the research of RA etiology and treatment. Auto-reactive CD4+ T cells are the main component in the etiology of RA/CIA, and inhibition of CD4+ T cells activition maight downregulate immune response specifically in RA. MiRNAs are important molcules regulating cell proliferation and differentiation. The role of miRNAs in CD4+T cells activation need to be investigated. In our prelimary study, miR-144 was found to be involved in the regulation of T cells in RA/CIA. In this study, we further analyze the expression of miRNAs in CIA CD4+ T cells, and detect the effects of the selected miRNAs in the proliferation and differentiation of T cells in CIA. The mechanism of miRNAs is further analyzed. The study provide experimental evidence for the role of miRNAs on RA etiology and new specific RA treatment method at the onset period of RA.
