中文摘要: 本课题以控制性超排卵交配小鼠为研究对象,运用补肾方,活血方以及补肾安胎方对该模型小鼠进行治疗,应用分子生物学、免疫组织化学、形态学定量方法以及扫描电镜等技术,采取动态、连续取样的方式,观察控制性超排卵对小鼠子宫内膜上VEGF 、Ang-1、2、PGI2 、PGE2、PGIS、PGES、PPARδMP2,MMP9,TIMP及其相关受体、子宫内膜形态学以及胞饮突的形成发育过程的影响,对补肾安胎方全方及其拆方的作用机制进行阐述。 本项目研究显示:补肾安胎方及其拆方均在改善控制性超排卵模型小鼠的着床方面有较好的作用,其中以补肾安胎全方组的作用优于两个拆方组(补肾组分、活血组分),表明两者存在协同作用,而活血方在促进血管生成方面比较有优势。其机制为:一,能够调节着床期间血清雌孕激素的含量以及雌孕激素受体在子宫内膜上的表达,有利于子宫内膜和胚胎间的对话;二,使着床局部血管通透性增强以及血管密度增加,Ang-1,2,VEGF均较模型组显著增强;三:能够调节小鼠子宫内膜着床局部MMP2,MMP9以及TIMP平衡,有利于着床期间子宫内膜蜕膜化。

英文摘要: The mouse model of controlled ovarian hyperstimulation was applied, then treated by Bushenan recipe, Huoxue recipe and Bushenantai recipe. We adopt molecular biology, immunohistochemistry, quantitative morphology and scanning electron microscope to investigate the mechanism of Bushen recipe, Huoxue recipe and Bushenantai Recipe ˊefffects on the implantation embryo dynamically and continuously. We observe the expression of Vascular endothelial growth factor(VEGF), Angiogenin-1,2, Prostacyclin I2, Prostacyclin E2, Prostacyclin I2 synthetase, Prostacyclin E2 synthetase, Peroxisome Proliferator-activated receptor, Matrix metalloproteinase-2,9, Tisse inhibitor of metalloproteinase and their interrelated receptors, pinopodes and the change of endometrial morphology. The project showed that Bushen recipe, Huoxue recipe and Bushenantai recipe can improve the implantation of embryo significantly, among which Bushenantai recipe is best, and Huexue recipe have advantage in angiogenesis. The mechanism is: First,they can regulate the contents of Estrogen, Progestogen and their receptors in blood-serum during the implantation of embryo, and profit the decidualization of endometrium and the dialogue between endometrium and embryo. Second, these recipes can increase the vasopermeability at the site of embryo implantation and the density of blood vessel. They improve the content of Ang-1, 2 and VEGF in endometrium dramatically. Third, they can adjust the balance of MMP2, MMP9 and TIMP in the implantation of endometrium, profit the decidualization of endometrium.
