中文摘要: 囿于明确、特异、可靠的阿尔茨海默病 (Alzheimer's disease,AD)早期临床诊断方法的缺乏,其已成为困扰医学界多年的难题,尤其是基于血液检测的生化指标的确立。本研究拟以AD病人及其正常对照组为研究对象,应用荧光关联光谱(FCS)及全内反射荧光显微(TIRFM)检测技术,在单分子水平上建立FCS和TIRFM定量检测AD病人血浆中Abeta42/Abeta40比值的超灵敏分析方法,并动态分析AD病人血浆中Abeta42/Abeta40的变化水平。项目的预期研究结果将进一步证实血浆中Abeta42/Abeta40作为AD生物学标记物的可靠性与特异性,不仅对AD早期临床诊断方法的确立具有重要意义,同时将对诸如肿瘤在内的其它重大疾病的诊断提供良好而积极的科学依据。

英文摘要: The lack of biomarkers has partly hampered the development of early diagnosis of Alzheimer disease (AD), especially for the blood biomarkers. In this study, we plan to investigate the plasma Abeta42/Abeta40 ratio in AD patients and matched healthy controls by using Fluorescence Correlation Spectroscopy (FCS) and Total Internal Reflection Fluorescence Microscopy (TIRFM). The dynamic analysis of plasma Abeta42/Abeta40 ratio in AD patients will also be performed by using the sensitive single molecule fluorescence technology. The reliability and specificity of the plasma Abeta42/Abeta40 ratio as a diagnostic tool for AD will be explored and this work will also open a new area for the diagnosis of other disorders such as tumor.
