中文摘要: 细胞迁移是细胞运动的基本形式之一,在肿瘤侵袭和转移过程中扮演关键角色;然而有关长链非编码RNA(LncRNA)在肿瘤细胞迁移运动中的作用及其分子机制知之甚少。我们应用生物芯片技术,同时筛选在胃癌组织中差异表达的LncRNA和mRNA,发现了多个胃癌相关的LncRNA,其中包括功能未知的LncRNA-LOC;结果显示LncRNA-LOC在胃癌组织中显著下调表达,改变其表达水平可明显影响肿瘤细胞的迁移能力。本课题将综合应用生物信息学、分子细胞生物学、生物化学及现代成像等技术,在分子、细胞、动物和临床等四个层面,系统研究LncRNA-LOC在肿瘤细胞恶性生物学行为(特别是肿瘤细胞的迁移和侵袭等)中的功能及其分子作用机制,并评估其与胃癌临床病理特征(尤其肿瘤侵袭与转移)的相关性。本课题的开展将有助于进一步了解肿瘤细胞迁移的分子调节机理,为肿瘤的诊断治疗提供新的理论依据和潜在治疗靶点。

英文摘要: Cell migration is one of the basic forms of cell motility, which plays a key role in the process of tumor invasion and metastasis. However, the function and mechanism of LncRNA in regulating the tumor cell migration and cell motility, hasn't yet been reported. We detected the expression profile of LncRNA and mRNA in gastric cancer samples compared with their adjacent normal gastric mucosa. Our microarray data identified a series of differentially expressed LncRNAs in gastric cancer. LncRNA-LOC,is one of the downregulated LncRNAs in the gastric cancer. Preliminary study showed that changing LncRNA-LOC expression in gastric cancer cell lines can greatly modulate cell migration ability. Based on these new findings, this project aims to study the role of LncRNA-LOC in regulating the biological behaviors of tumor cells, especially cell migration and cell invasion, through kinds of modern technology of bioinformatics, molecular cytobiology, biochemistry and imaging.Moreover, we will study this project at four different aspects including molecular level, cellular level, animal level, and clinical level. The launching of this study will help to further understand the molecular regulation mechanism of tumor cell migration, and provides a new potential therapeutic targets for the diagnosis and treatment of gastric cancer.
