中文摘要: 随着核技术在军事和民事领域广泛应用,急性放射病研究成为重要的医学急救知识储备。其中极重度骨髓型放射病目前世界无存活案例,成功的造血干细胞移植可以使该类患者造血快速恢复,但无法长期存活,主要屏障是免疫系统受损和多组织脏器射线损伤,组织细胞凋亡坏死,最终导致多脏器衰竭死亡。本课题组既往工作证明标记的骨髓间充质干细胞MSC输注致死剂量照射小鼠后,在胸腺组织见标记的MSC归巢并大量扩增,同时发现其与骨髓细胞共移植促进小鼠CD4细胞早期增值,与既往文献报道的MSC免疫抑制作用不同,因此,本课题组准备在以往工作基础上,进一步探讨MSC促进射线损失的胸腺组织修复和免疫功能恢复的作用机理,并在此研究结果基础上,用KGF和IL-7对MSC进行基因转染,从而加强MSC对受照胸腺的组织修复和免疫功能恢复作用,为临床救治极重度放射病患者,提高存活率,开辟新的治疗措施。

英文摘要: With nuclear technique carrying out in military and civil fields the study of acute radiation syndrome(ARS) become the most emergency medical knowledge reserve.The patients with severe bone marrow tape ARS had no long term survival.The hematopoietic stem cells transplantation made these patients rapid hamatopoietic recovery after radiation exposure, but immune system injury and tissure injury made multiple organ failture to patients final death.Our last experiment study showed that mesenchymal stem cells can found large expansion in thymus after infusion lethal dose radiation mice,and MSC combined with hematopoietic stem cells transplantation can improve CD4 cells early recovery.So our aim is deep study on the machinism of MSC improving mythus and immnue system recovery, furthermore KGF and IL-7 transfact MSC to enhance repair function.Based on above experiments we hope to make a new measure to improve survival rate for severe ARS patients.
