中文摘要: 课题组前期通过整合基因组学与血清蛋白质组学(SELDI方法),研究发现调节肿瘤细胞增殖和凋亡平衡的细胞色素C1蛋白(CYC1)在骨肉瘤患者血清中表达显著增强且表达程度与机体荷瘤量呈正相关,提示该蛋白是潜在的骨肉瘤血清标志蛋白。目前我们已构建CYC1转基因骨肉瘤细胞株并正在构建原位骨肉瘤裸鼠模型,拟进一步探索CYC1对骨肉瘤生物学性状的在体调节作用,并确认CYC1是否在细胞和动物水平上通过激活凋亡通路中Caspases、Bcl-2、Fas等效应因子来调节骨肉瘤细胞的凋亡,研究凋亡信号通路上CYC1相关蛋白表达。通过本研究,我们希望能从细胞和动物整体水平深入了解CYC1对骨肉瘤细胞凋亡的调节机制,进一步揭示CYC1的生物学功能及其对骨肉瘤恶性生物学行为的影响,为CYC1作为骨肉瘤临床诊断血清学标志物以及基因治疗靶点提供理论依据。

英文摘要: In the previous research of our project, with the Surface Enhanced Laser Desorption/Ionization-Time of Flight-Mass Spectrometry (SELDI-TOF-MS) method, we have found that Cytochrome C1 (CYC1), which plays an important role in the balance of cell proliferation and apoptosis, has a higher expression in osteosarcoma patient's serum and the quantity of CYC1 is positively correlated with the tumor size, which suggests that the expression of CYC1 can be used as the sign of osteosarcoma. We have constructed the CYC1 transgenetic cell line of osteosarcoma and are trying to raise nude mouse model of osteosarcoma in situ using these cells in order to study the function of CYC1 in osteosarcoma patient's body and confirm whether CYC1 can activate Caspases, Bcl-2 and Fas to adjust the apoptosis of cell, which will provide sufficient basis for the diagnosis of osteosarcoma and targeted therapy.
