中文摘要: 磁性纳米粒子在生物医用领域具有重要的应用前景,但如何得到能高效运载基因药物和化学药物,对药物达到可控释放,超灵敏识别癌细胞,诊断癌症以及报告癌细胞的死亡等功能为一体的智能型磁性纳米药物载体,仍是当前面临的一大挑战。 本项目拟首先设计合成多孔空心金-四氧化三铁纳米粒子,然后分别将氧化石墨烯、靶向肝癌细胞的iRGD肽和稀土上转换纳米粒子通过功能化修饰在其表面,金纳米粒子内装化学药物,氧化石墨烯表面负载基因药物,构建出智能型的基因-化学药物共传输磁性纳米复合控释载体。这种载体可实现靶向识别肝癌细胞,无损原位诊断肝癌,可控释放药物,联合治疗肝癌并可视化实时监测治疗过程, 从而根本上改变肿瘤学诊断、治疗和预防肝癌的方式。 此项目的完成,将为纳米药物控释系统的研究和临床实验提供基础数据,并为今后多模式诊断和治疗癌症提供重要科学依据。

英文摘要: The magnetic nanoparticles in the biomedical field have an important application prospect.But there is still a challenge about how to obtaine intelligent magnetic nanoparticles as drug carriers, which could simultaneously efficiently deliver chemical drugs and gene drugs, control the gene therapeutic drugs and chemical drugs release in tumor cells, super-sensitive recognize the cancer cells, diagnose cancer, report cancer cells death. This project aims to construct intelligent magnetic nano composite material as gene-chemical drug co-delivery carries. Fristly, the porous hollow Au-Fe3O4 nanoparticles will be designed and synthesized, and then Graphene oxide, Rare earth conversion nanoparticles and iRGD pepite targeting to liver cancer cells throught fuction will be modified on the surface of porous hollow Au-Fe3O4 nanoparticles. The chemical drugs are loaded in the hollow gold nanoparticles, and gene drugs are carried on the surface of Graphene oxide. This carrier could realize target identification of liver cancer cells, nondestructive in situ diagnosis of liver cancer, controlled release of drugs, combined therapy of liver cancer and real-time monitoring of the treatment process with visualization.
