中文摘要: 牙釉质再生是牙再生研究中的难点。由于成熟牙釉质不含细胞成分,因此常规组织工程技术难以实现牙釉质组织再生。利用人工合成基质仿生牙釉质形成过程中的基质蛋白,引导成釉细胞在人工基质上分泌,以促进牙釉质的新生,是探索牙釉质再生的新方法。本课题计划在球形PAMAM分子上接枝RGD和釉原蛋白功能序列LRAP的末端肽,构建可促进牙釉质新生的人工基质,可以利用精确控制的球形PAMAM分子结构,实现对新生牙釉质形成的控制。通过对新生牙釉质进行microCT、组织学观察,对晶体进行检测,反馈调整人工基质分子结构,最终获得可调控牙釉质新生的人工基质。课题组具有球形PAMAM分子促进牙釉质仿生矿化等研究基础,实验条件具备。本课题将有望为牙釉质再生提供仿生人工基质,也可以为探索牙釉质发育过程中细胞外基质的作用提供研究方法。

英文摘要: he regeneration of dental enamel occupies the hardest part of tooth regeneration. Ordinary tissue engineering methods were proved to be in vain. A new method was raised by this study that is to utilize artificial matrix to biomimic the extracellular matrix in amelogenesis which have been verified to conduct ameloblast excretion and following amelogenesis. As artificial matrix, in this study, sphere-shaped PAMAM dendrimers are planned to synthesized and decorated on the outer ends by RGD and functional peptide from amlogenin's LRAP (Leucine-rich Amelogenin Peptide). The functionalized dendrimers have precise structures which can be adjusted accurately. So, manipulating of the artificial molecular may provide an approach to control the amelogenesis. According to the investigation of the novel enamel generation by microCT and histological methods, the structure of the sphere-shaped PAMAM should be calibrated for better dental enamel growth. Finally, an artificial matrix which can control the dental enamel growth will be obtained. Our research group have many basic research works such as sphere-shaped PAMAM which can promote biomimic dental enamel mineralization. In this study, the artificial matrix, which will be developed, may act as bio-template for enamel regeneration in future, and also a new method to investigate the function of extracellular matrix in amelogenesis.
