中文摘要: 黏膜免疫系统是机体抗感染的第一道防线,在抵抗感染方面起着极其重要的作用。硒作为人和动物生长发育必需的微量元素,其缺乏对肠粘膜免疫系统产生重要的影响。本项目拟以雏鸡为试验动物,建立缺硒雏鸡模型,运用2D-DIGE、质谱鉴定、生物信息学分析等技术分离筛选缺硒雏鸡与健康雏鸡肠黏膜系统差异表达的蛋白,利用荧光定量PCR和Western blot方法分别从基因和蛋白水平对差异表达蛋白进行进一步验证,然后对差异表达蛋白分类后进行功能预测分析,揭示硒缺乏对雏鸡肠黏膜系统功能影响的分子机制,为硒在肠黏膜系统中的作用机制提供理论依据。

英文摘要: The mucosal immune system is the first defense that the body use to fight infection, which plays an extremely important role in resisting infection. Selenium is the necessary trace elements in the growth and development of humen and animals, selenium deficiency of bowel mucosa immune system has the important influence. For the project, we intend to use chicks for the experimental animals. A model is established in which chicks lack selenium. The screening methods 2D-DIGE, mass spectrum identification, and bioinformatics analysis technology are used to screen the differential expression proteins in intestinal mucosa of selenium deficiency chicks and healthy chickens. The differential expression proteins were verified at mRNA and protein levels using quantitative PCR and Western blot methods. Furthermore, the differential proteins were classified on basis of function, and the function involving selenium regulation was prediced. Finally, we reveal the molecular mechanisms that effect of selenium deficiency on the chick intestinal mucosal system functions, and provide some basis for understanding selenium function in intestinal mucosal system.
