中文摘要: 目前研究发现细胞免疫途径亦是膜性肾病的发病机制。本项目是在前期以调节细胞免疫功能干预MN动物模型研究的基础上,以CD4+ CD25+ Treg细胞为研究切入点,从调控CD4+ CD25+ Treg细胞干预GBM病变这一视角,结合MN的病机关键-少阳枢机不利,肾络郁滞,提出疏利少阳,辛通畅络治法,以临床疗效显著的肾络宁为研究载体,围绕 疏利少阳,辛通畅络法通过调控CD4+ CD25+Treg细胞,抑制中性粒细胞分泌MMP-9,改善GBM的IV型胶原病变是阻抑MN的有效途径这一假说,采用细胞剔除、基因敲除、流式细胞仪、透射电镜、免疫组化、RT-PCR等多层面技术,动态观察CD4+ CD25+ Treg细胞与中性粒细胞、MMP-9和IV型胶原表达的相关性,揭示细胞免疫在MN发病机制中的作用,同时探讨中医药通过调控CD4+ CD25+ Treg细胞,改善GBM病变的可能性及深层机制。

英文摘要: Recently research discovery that cellular immunity path is a significance pathogenesises of membranous nephropathy(MN). Based on before research that treat MN Animal Models by regulate cellular immune function, this study raise the theory that dysfunction of Shaoyang and emphraxis of kidney collaterals is pathogenesis of MN and therapy that regulate Shaoyang and deoppilation kidney collaterals,in order to prove the hypothesis that Shenluoning could treat pathological changes of glomerular basement membrane(GBM) by regulate CD4+ CD25+ Treg cells and suppress neutrophil secrete matrix metalloproteinase-9(MMP-9). So this study adopt cell reject, gene knockout, flow cytometer, electron microscope, immunohistochemisty, RT-PCR to reveal the correlation of CD4+ CD25+ Treg cells with neutrophil, MMP-9 and collagen Ⅳ of GBM, reveal the mechanism of cellular immunity in MN,approach probability that treat pathological changes of GBM by regulate CD4+ CD25+ Treg cells with TCM.
