中文摘要: microRNA作为一类重要的非编码调控分子,在调节胚胎干细胞(ES)自我更新及分化以及诱导性多能干细胞(iPS)产生过程中发挥着重要作用。我们研究发现miR-221~222簇在小鼠ES细胞多种分化模式中均表达上调,且该miRNA簇成员在小鼠ES细胞中的过表达均能明显地抑制ES细胞的自我更新,促进ES细胞向各胚层分化。此外我们鉴定出多个自我更新相关转录因子及关键信号通路分子均为miR-221~222簇的下游靶基因。在以上基础上,本项目将进一步深入研究miR-221~222簇调节小鼠ES细胞分化的功能和机制。且因为miR-221~222簇在胚胎成纤维细胞(MEF)中含量非常丰富,我们将在使用MEF诱导多能干细胞产生过程中抑制该miRNA簇的表达,检测其对于iPS效率及质量的影响。这些工作将丰富miRNA在胚胎干细胞领域的研究,并有助于指导胚胎干细胞的体外定向分化,有助于优化现有的iPS技术。

英文摘要: MicroRNAs (miRNAs) have emerged as critical regulators of embryonic stem cells (ES) and induced pluripotent stem cells (iPS) by working at the post-transcriptional level. MiR-221~222 cluster is upregulated during RA-induced embryonic stem cell differentiation and EB formation. Enforced expression of miR-221,222 can suppress the self-renewal in mES cells and meanwhile promote mES differentiation. We also found that miR-221~222 cluster regulates mES differentiation through targeting several transcriptional factors and multiple members of signal pathway responsible for mES self-renewal. We will further study the function and mechanism of miR-221~222 cluster modulating mES differentiation. Since miR-221~222 cluster is enriched in mouse embryonic fibroblast(MEF),we will try to improve the efficiency and pluripotency of iPS cells through suppressing the expression of miR-221~222 cluster during generation of iPS cells from MEF.These works are useful for the committed differentiation of ES cells and the generation of iPS cells.
